    POST-IRRADIATION ANALYSES OF U-MO DISPERSION FUEL RODS OF KOMO TESTS AT HANARO

    Since 2001, a series of five irradiation test campaigns for atomized U-Mo dispersion fuel rods, KOMO-1, -2, -3, -4, and -5, has been conducted at HANARO (Korea) in order to develop high performance low enriched uranium dispersion fuel for research reactors. The KOMO irradiation tests provided valuable information on the irradiation behavior of U-Mo fuel that results from the distinct fuel design and irradiation conditions of the rod fuel for HANARO. Full size U-Mo dispersion fuel rods of 4–5 g-U/cm3 were irradiated at a maximum linear power of approximately 105 kW/m up to 85% of the initial U-235 depletion burnup without breakaway swelling or fuel cladding failure. Electron probe microanalyses of the irradiated samples showed localized distribution of the silicon that was added in the matrix during fuel fabrication and confirmed its beneficial effect on interaction layer growth during irradiation. The modifications of U-Mo fuel particles by the addition of a ternary alloying element (Ti or Zr), additional protective coatings (silicide or nitride), and the use of larger fuel particles resulted in significantly reduced interaction layers between fuel particles and Al

    Background: Although the relation of the gut microbiota to a development of autoimmune and inflammatory diseases has been investigated in various animal models, there are limited studies that evaluate the effect of probiotics in the autoimmune eye disease. Therefore, we aimed to investigate the effect of IRT-5 probiotics consisting of Lactobacillus casei, Lactobacillus acidophilus, Lactobacillus reuteri, Bifidobacterium bifidum, and Streptococcus thermophilus on the autoimmunity of uveitis and dry eye and alloimmunity of corneal transplantation. Methods: Experimental autoimmune uveitis was induced by subcutaneous immunization with interphotoreceptor-binding protein and intraperitoneal injection of pertussis toxin in C57BL/6 (B6) mice. For an autoimmune dry eye model, 12-weeks-old NOD.B10.H2b mice were used. Donor cornea of B6 mice was transplanted into BALB/C mice. IRT-5 probiotics or phosphate buffered saline (PBS) were administered for three weeks immediately after induction of uveitis or transplantation. The inflammation score of the retinal tissues, dry eye manifestations (corneal staining and tear secretion), and graft survival were measured in each model. The changes of T cells were evaluated in drainage lymph nodes using fluorescence-activated cell sorting. Results: Retinal histology score in IRT-5 group of uveitis was lower than that in PBS group (p = 0.045). Ocular staining score was lower (p < 0.0001) and tear secretion was higher (p < 0.0001) in the IRT-5 group of NOD.B10.H2b mice than that in the PBS group. However, the graft survival in the IRT-5 group was not different from those of PBS group. The percentage of regulatory T cells was increased in the IRT-5-treated dry eye models (p = 0.032). The percentage of CD8+IL-17hi (p = 0.027) and CD8+ interferon gamma (IFN��)hi cells (p = 0.022) were significantly decreased in the IRT-5-treated uveitis models and the percentage of CD8+IFN��hi cells was markedly reduced (p = 0.036) in IRT-5-treated dry eye model. Conclusion: Our results suggest that administration of IRT-5 probiotics may modulate clinical manifestations of autoimmunity in the eye, but not on alloimmunity of corneal transplantation. ? 2017 by the authors.112Nsciescopu

    Clinical and genomic assessment of PD-L1 SP142 expression in triple-negative breast cancer

    Purpose: The SP142 PD-L1 assay is a companion diagnostic for atezolizumab in metastatic triple-negative breast cancer (TNBC). We strove to understand the biological, genomic, and clinical characteristics associated with SP142 PD-L1 positivity in TNBC patients. Methods: Using 149 TNBC formalin-fixed paraffin-embedded tumor samples, tissue microarray (TMA) and gene expression microarrays were performed in parallel. The VENTANA SP142 assay was used to identify PD-L1 expression from TMA slides. We next generated a gene signature reflective of SP142 status and evaluated signature distribution according to TNBCtype and PAM50 subtypes. A SP142 gene expression signature was identified and was biologically and clinically evaluated on the TNBCs of TCGA, other cohorts, and on other malignancies treated with immune checkpoint inhibitors (ICI). Results: Using SP142, 28.9% of samples were PD-L1 protein positive. The SP142 PD-L1-positive TNBC had higher CD8+ T cell percentage, stromal tumor-infiltrating lymphocyte levels, and higher rate of the immunomodulatory TNBCtype compared to PD-L1-negative samples. The recurrence-free survival was prolonged in PD-L1-positive TNBC. The SP142-guided gene expression signature consisted of 94 immune-related genes. The SP142 signature was associated with a higher pathologic complete response rate and better survival in multiple TNBC cohorts. In the TNBC of TCGA, this signature was correlated with lymphocyte-infiltrating signature scores, but not with tumor mutational burden or total neoantigen count. In other malignancies treated with ICIs, the SP142 genomic signature was associated with improved response and survival. Conclusions: We provide multi-faceted evidence that SP142 PDL1-positive TNBC have immuno-genomic features characterized as highly lymphocyte-infiltrated and a relatively favorable survival

    We discuss three seemingly unrelated quantities that have been introduced in different fields of science for complex networks. The three quantities are the resistance distance, the information centrality and the node displacement. We first prove various relations among them. Then we focus on the node displacement, showing its usefulness as an index of node vulnerability.We argue that the node displacement has a better resolution as a measure of node vulnerability than the degree and the information centrality

    Genome sequence of Jatropha curcas L., a non-edible biodiesel plant, provides a resource to improve seed-related traits

    Jatropha curcas (physic nut), a non‐edible oilseed crop, represents one of the most promising alternative energy sources due to its high seed oil content, rapid growth and adaptability to various environments. We report ~339 Mbp draft whole genome sequence of J. curcas var. Chai Nat using both the PacBio and Illumina sequencing platforms. We identified and categorized differentially expressed genes related to biosynthesis of lipid and toxic compound among four stages of seed development. Triacylglycerol (TAG), the major component of seed storage oil, is mainly synthesized by phospholipid:diacylglycerol acyltransferase in Jatropha, and continuous high expression of homologs of oleosin over seed development contributes to accumulation of high level of oil in kernels by preventing the breakdown of TAG. A physical cluster of genes for diterpenoid biosynthetic enzymes, including casbene synthases highly responsible for a toxic compound, phorbol ester, in seed cake, was syntenically highly conserved between Jatropha and castor bean. Transcriptomic analysis of female and male flowers revealed the up‐regulation of a dozen family of TFs in female flower. Additionally, we constructed a robust species tree enabling estimation of divergence times among nine Jatropha species and five commercial crops in Malpighiales order. Our results will help researchers and breeders increase energy efficiency of this important oil seed crop by improving yield and oil content, and eliminating toxic compound in seed cake for animal feed

    Transition from in-plane to out-of-plane azimuthal enhancement in Au+Au collisions

    The incident energy at which the azimuthal distributions in semi-central heavy ion collisions change from in-plane to out-of-plane enhancement, E_tran, is studied as a function of mass of emitted particles, their transverse momentum and centrality for Au+Au collisions. The analysis is performed in a reference frame rotated with the sidewards flow angle, Theta_flow, relative to the beam axis. A systematic decrease of E_tran as function of mass of the reaction products, their transverse momentum and collision centrality is evidenced. The predictions of a microscopic transport model (IQMD) are compared with the experimental results.Comment: 32 pages, Latex, 22 eps figures, accepted for publication in Nucl. Phys.

    Slewing Mirror Telescope and the Data-Acquisition System for the UFFO-Pathfinder

    The Ultra-Fast Flash Observatory (UFFO) aims to detect the earliest moment of Gamma-Ray Bursts (GRBs) which is not well known, resulting into the enhancement of GRB mechanism understanding. The pathfinder mission was proposed to be a scaled-down version of UFFO, and only contains the UFFO Burst Alert & Trigger Telescope (UBAT) measuring the X-ray/gamma-ray with the wide-field of view and the Slewing Mirror Telescope (SMT) with a rapid-response for the UV/optical photons. Once the UBAT detects a GRB candidate with the position accuracy of 10 arcmin, the SMT steers the UV/optical photons from the candidate to the telescope by the fast rotatable mirror and provides the early UV/optical photons measurements with 4 arcsec accuracy. The SMT has a modified Ritchey-Chrètien telescope with the aperture size of 10 cm diameter including the rotatable mirror and the image readout by the intensified charge-coupled device. There is a key board called the UFFO Data Acquisition system (UDAQ) that manages the communication of each telescope and also of the satellite and the UFFO overall operation. This pathfinder is designed and built within the limited size and weight of  ~20 kg and the low power consumption up to  ~30 W. We will discuss the design and performance of the UFFO-pathfinder, and its integration to the Lomonosov satellite

    Design and implementation of electronics and data acquisition system for Ultra-Fast Flash Observatory

    The Ultra-Fast Flash Observatory (UFFO) Pathfinder for Gamma-Ray Bursts (GRBs) consists of two telescopes. The UFFO Burst Alert & Trigger Telescope (UBAT) handles the detection and localization of GRBs, and the Slewing Mirror Telescope (SMT) conducts the measurement of the UV/optical afterglow. UBAT is equipped with an X-ray detector, analog and digital signal readout electronics that detects X-rays from GRBs and determines the location. SMT is equipped with a stepping motor and the associated electronics to rotate the slewing mirror targeting the GRBs identified by UBAT. First the slewing mirror points to a GRB, then SMT obtains the optical image of the GRB using the intensified CCD and its readout electronics. The UFFO Data Acquisition system (UDAQ) is responsible for the overall function and operation of the observatory and the communication with the satellite main processor. In this paper we present the design and implementation of the electronics of UBAT and SMT as well as the architecture and implementation of UDAQ
